SO, if I build my quary, build a level 20 wall and then destroy my quary, will my wall still exist and defend me?

only need a level one Stonemason to do the Research Defensive Structure which is the thing that lets you build the wall so yes it will still exist and defend you

OK, so, do I need to maintain a quary while I build my wall up, or do I just need it long enough to complete the research?

I am pretty sure as soon as you start the you research you no longer Required building (Stonemason ) anways shouldnt demo the Quarry

Demolishing quarries will not increase the number of building spaces available in your city. You cannot build other city buildings on resource spots such as quarries, lumberyards, iron mines, clay pits or farms.

These resource fields consume very low amounts of food (maximum of 37 per hour at level 20). Therefore there is little advantage in demolishing them.
